PhD position (Molecular Biology): RNA mediated regulation of gene expression

In the field of “RNA mediated regulation of gene expression” the research group of Dr Jennifer Winter offers the following PhD project:

Non-coding RNA profiles in sperm of resilient and susceptible animals


Our hypothesis-free approach will apply RNAseq and RT-qPCR and mainly focus on male germ cells-delivered small noncoding (snc) and long noncoding (lnc) RNAs and RNA-editing modifications as epigenetic factors that transmit acquired phenotypes. Snc- and lncRNAs upregulated in sperm of R+ and R- mice will be functionally tested for their ability to directly induce enhanced or reduced resilience. To this end, we will perform microinjections of these ncRNAs into wild-type (wt) zygotes and analyze the resulting offspring regarding phenotypical similarity to R+ and R- offspring of male R+ and R- CSD mice.

The project addresses the following questions: Are CSD experiences (e.g. sensed by the brain of the parent) and altered resilience capacity translated into epigenetic changes in the germ cells and transmitted to subsequent generations? Clarifying the precise time point of the transition from the original ncRNA response to a stable epigenetic mechanism based on the genomic DNA level that could be maintained during mitosis and germ cell development. What is the role of post-transcriptional RNA modifications in transgenerational epigenetic inheritance?
